I am Chengfu Xu, MD, Professor, Chief Physician and Deputy Director, Department of Gastroenterology, Zhejiang Provincial Clinical Research Centre for Digestive Diseases, the First Affiliated Hospital, Zhejiang University School of Medicine. My research mainly focuses on exploring the pathogenesis and interventions of metabolic dysfunction associated fatty liver disease (MAFLD). Metabolic dysfunction associated fatty liver disease is one of the four major chronic diseases in China, and is prone to progress to liver fibrosis, cirrhosis, and even liver cancer. Resmetirom is currently the only therapeutic drug for metabolic dysfunction associated fatty liver disease, but its effective rate is less than 30%, and the fundamental reason is that the pathogenesis of metabolic dysfunction associated fatty liver disease is largely unknown. My previous work includes: (1) Establishment of a large-scale metabolic dysfunction associated fatty liver disease cohort, which for the first time demonstrated that hyperuricemia significantly increases the risk of metabolic dysfunction associated fatty liver disease onset and long-term adverse outcomes; (2) Elucidation of novel mechanisms by which hyperuricemia promotes metabolic dysfunction associated fatty liver disease progression, including NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome activation and mitochondrial damage in hepatocytes; and (3) Discovery of the beneficial effects of xanthine oxidase-targeted urate-lowering interventions in metabolic dysfunction associated fatty liver disease treatment and prevention. These results were published in Hepatology, Journal of Hepatology, Nature Communications, Cellular and Molecular Gastroenterology and Hepatology, Journal of Biological Chemistry, etc. Notably, the research outcomes have been incorporated into the textbook "Internal Medicine" and international and domestic guidelines for fatty liver disease diagnosis and treatment. I have been honored with the First Prize of Scientific and Technological Progress Award by the Ministry of Education (ranked 3/13), the Second Prize of China Medical Science and Technology Award (ranked 3/10), and the Second Prize of Zhejiang Provincial Scientific and Technological Progress Award (ranked 2/10 and 6/11).
