Abstract
BACKGROUND Evidence of dual sensory impairment (DSI) characterized by comorbid hearing impairment (HI) and visual impairment (VI) is limited in neuropsychiatric disorders. Moreover, the underlying mechanisms remain elusive. METHODS Using data from 169,165 UK Biobank participants, we compared cognition and mental health across the no sensory impairment, HI, VI, and DSI groups and investigated the association between the number of sensory impairments and neuropsychiatric disorders in longitudinal COX analyses. We also analyzed the associations of sensory function and its interaction with brain structure, functional connectivity, inflammatory markers, and metabolites and explored their mediating roles. RESULTS DSI participants exhibited poor cognition and mental health, and had a higher risk of neuropsychiatric disorders compared to those with HI/VI alone. Both HI and VI were linked to atrophic changes in the hippocampus, thalamus, precuneus, parietal cortex, middle temporal gyrus, etc., and reduced white matter integrity. Brain regions associated with HI and VI exhibited decreased functional connectivity with other cortexes. Hearing and visual performance exhibit a significant interaction in relation to cognitive function, mental health, and metabolites. Brain structures, inflammation, and metabolic biomarkers significantly associated with HI and VI mediated the relationship between sensory impairments and cognition/mental health. CONCLUSION Compared to HI/VI alone, DSI is associated with poorer cognitive function, mental health, and a higher risk of neuropsychiatric disorders, potentially driven by the interaction between HI and VI. Additionally, HI and VI are linked to brain atrophy, reduced white matter integrity, and altered functional connectivity. Furthermore, brain structure, inflammation, and metabolic biomarkers mediate the relationship between sensory impairments and cognition/mental health.

Abstract
Insomnia affects a substantial proportion of the population and frequently co-occurs with mental illnesses including depression and anxiety. However, the neurobiological correlates of these disorders remain unclear. Here we review magnetic resonance imaging (MRI) studies assessing structural and functional brain associations with depressive and anxiety symptoms in insomnia disorder (ID; n = 38), insomnia symptoms in depressive and anxiety disorders (n = 14), and these symptoms in the general populations (n = 3). The studies on insomnia disorder consistently showed overlapping (salience network: insula and anterior cingulate cortex) and differential MRI correlation patterns between depressive (thalamus, orbitofrontal cortex and its associated functional connectivity) and anxiety (functional connectivity associated with default mode network) symptoms. The insula was also consistently identified as indicating the severity of insomnia symptoms in depressive disorder. In contrast, findings for other regions related to insomnia symptoms in both depressive and anxiety disorders were generally inconsistent across studies, partly due to variations in methods and patient cohorts. In the general population, brain regions in the default mode network provided a functional link between insomnia and depressive symptoms. These findings underscore both the shared and distinct neural correlates among depression, anxiety, and insomnia, providing potential avenues for the clinical management of these conditions.

Abstract
BACKGROUND AND OBJECTIVES The association between biological aging and dementia, as well as the underlying mechanism, remains limited. The aim of this study was to investigate the relationships of biological age (BA) with incident dementia and examine the underlying neurobiological mechanisms. METHODS This study used data from the UK Biobank, a prospective longitudinal study. We included participants free of diagnosed dementia at baseline. BA was evaluated from clinical traits using the Klemera-Doubal method biological age (KDM-BA) and PhenoAge algorithms. Genetic risk of dementia was assessed using the apolipoprotein E (APOE) ε4 genotype and polygenic risk scores (PRSs). Cox proportional hazard regression models were used to estimate the associations of BA and the combined effect of genetic risk and BA with dementia. In addition, the potential roles of brain structures (gray matter volume [GMV], cortical mean thickness, and cortical surface area) in the associations between BA and dementia were evaluated using mediation analysis. RESULTS A total of 280,918 participants (mean age 56.80 years, 54.59% female) were enrolled in this study. Over a median follow-up of 13.58 years, 4,770 cases of dementia were recorded. Every SD increase in KDM-BA accelerations and PhenoAge accelerations was associated with a 14% (hazard ratio [HR] = 1.14; 95% CI 1.10-1.18) and 15% (HR = 1.15; 95% CI 1.12-1.19) higher incidence of dementia, respectively. Individuals with APOE ε4 and highest PhenoAge accelerations had the highest risk of dementia (HR = 4.20, 95% CI 3.69-4.78) compared with those with non-APOE ε4 and lowest PhenoAge accelerations, with significant interaction effect (Pinteraction < 0.001). We did not find significant modification effects of PRS on the associations between BA accelerations and dementia (Pinteraction = 0.347 for KDM-BA acceleration, Pinteraction = 0.279 for PhenoAge acceleration), as well as APOE ε4 on the association between KDM-BA accelerations and dementia (Pinteraction = 0.212). Mediation analysis showed that the identified GMV, cortical mean thickness, and cortical surface area partly mediated the association between BA accelerations and incident dementia, with proportion-mediated percentage ranging from 6.64% to 17.98%. DISCUSSION Advanced BA may be a potential risk factor of incident dementia. The risk is possibly mediated through the widespread reduction of brain structures.

Abstract
BACKGROUND Grounded in the 24-Hour Activity Cycle framework, this study aimed to quantify dose-response thresholds for seven daily behaviors-sleep, napping, low to high intensity physical activity, sedentary time, and screen time-and to evaluate their associations with mental health problems and self-injurious thoughts or behaviors in Chinese adolescents. METHODS Between 2022 and 2023, data were collected from 33,397 Chinese students to examine lifestyle patterns and mental health. Adjusted logistic regression estimated associations between behavioral categories and four mental-health outcomes, and generalized additive models captured dose-response relationships. RESULTS Compared with non-nappers, adolescents who napped 60-90 min daily had 16 % lower odds of suicidal risk (OR 0.84; 95 % CI 0.73-0.97), while moderate-intensity exercise on at least two days per week was associated with 12-31 % reduced odds across all mental health outcomes (OR range 0.69-0.88). Conversely, sleep under 7 h, daily sedentary time over 7 h, and screen time exceeding 4 h were linked to higher risk of adverse outcomes. Generalized Additive Models confirmed significant nonlinear dose-response relationships and identified a common set of healthy lifestyle ranges across all four outcomes: 7.5-10.5 h of sleep, over 40 min of napping, <7.5 h of sedentary time, and under 4.4 h of screen time per day. CONCLUSIONS These results provide critical evidence for designing policies and school-based programs aimed at fostering healthier daily routines, ultimately promoting the mental health and resilience of children and adolescents.

Abstract
BACKGROUND Percheron acute artery occlusion is a rare type of acute cerebral infarction. CASE SUMMARY An elderly male presented with sudden-onset near-memory and sensory impairments for 5 days. Upon admission, based on symptoms, signs, magnetic resonance imaging, and computed tomography findings, a diagnosis of Percheron syndrome was made. Subsequently, anti-platelet therapy, lipid-lowering treatment, cerebral circulation enhancement (such as the administration of calcium channel blockers to improve cerebral blood flow), and neurotrophic support (such as the use of drugs like citicoline to protect nerve cells) were immediately implemented, along with additional symptomatic treatments. The patient's symptoms were alleviated, following which he was discharged. CONCLUSION The diagnosis of acute occlusion of the Percheron artery requires rich clinical expertise and accurate imaging tools. Timely intervention and effective follow-up hold significant implications for optimizing patient recovery.

Abstract
BACKGROUND With the rapid aging of populations, dementia has become a major health problem. Hearing loss is common among older adults and has been recognized as a contributing factor to cognitive impairment and dementia. We aimed to explore the association between hearing loss and cognitive impairment in older adults. We also sought to quantify the extent to which the impact of hearing loss on cognitive impairment could be mediated by depression and social isolation. METHODS We used nationally representative data from the China Health and Retirement Longitudinal Study (CHARLS), a population-based cohort study. The study population comprised adults aged 60 and older who participated in at least two waves of the CHARLS. Cognitive function was measured based on four dimensions: orientation, computation, memory, and drawing. Hearing loss was defined according to self-reported data. We used logistic regression models to analyze the cross-sectional association between hearing loss and cognitive impairment. Cox proportional hazards models were used to examine the longitudinal association between hearing loss and cognitive impairment. We also conducted an in-depth analysis of the severity of hearing loss and its associated risk of cognitive impairment. Additionally, we calculated the percentage of excess risk mediated (PERM) by depression and social isolation. FINDINGS In total, 7891 individuals were included in the cross-sectional analysis, and 5326 were included in the longitudinal analysis, with a mean follow-up of 6.38 years. In the cross-sectional analysis, hearing loss was associated with cognitive impairment after adjusting for all covariates (OR = 1.76, 95% CI: 1.49-2.08). In the longitudinal analysis, the hazard ratio for cognitive impairment with hearing loss compared with no hearing loss was 1.24 (95% CI: 1.06-1.46) after adjustment for demographics, socioeconomic factors, and health-related behaviors. Further analysis revealed that moderate or severe hearing loss had a strong relationship with cognitive impairment after adjusting for covariates (HR = 1.32, 95% CI: 1.07-1.62). Of the total association between hearing loss and cognitive impairment, 20.83% was mediated by depression and 4.17% by social isolation. CONCLUSIONS Hearing loss is independently associated with cognitive impairment, providing evidence that addressing hearing loss could help improve cognitive function.

Abstract
Poor sleep is a known risk factor of cognitive disorders, but the role of individual sleep problems in age-related cognitive changes remains unclear. This study used two complementary statistical models to estimate nonlinear trajectories of decline in four domains of cognitive functioning in the age period between 55 and 100 years depending on the severity of problems with falling asleep, night awakenings, and early morning awakenings, and short/long sleep duration. The sample included 5132 older adults (M = 67 years, 48% male) from the Longitudinal Aging Study Amsterdam (LASA), assessed 4-10 times every 2-3 years. Sleep problems were self-reported, and cognitive functioning was measured with the 15-Word test (reflecting episodic memory as immediate and delayed recall), Coding task (information processing speed) and Mini-mental State Examination/MMSE (global cognition). Data were analysed using quadratic and piecewise changepoint mixed models. The piecewise models provided more precise and interpretable findings. Decline in information processing speed accelerated significantly earlier in participants with short sleep duration (regression coefficient (B) = -2.3[95% confidence interval (CI): -3.86; -0.81]; p < 0.01) and faster with more severe early morning awakenings (B = -0.07 [-0.1; -0.03]; p < 0.01). Decline in immediate recall accelerated earlier in those with short sleep (B = -2.8 [-4.44; -1.14]; p < 0.01) and severe problems with falling asleep (B = -1.22 [-2.06; -0.39]; p = 0.01). Decline in delayed recall was faster with long sleep (B = -0.06 [-0.08;-0.03]; p < 0.01). Decline in global cognition accelerated faster in those with short/long sleep duration (B = -0.07 [-0.13; -0.01]/-0.10 [-0.18; -0.03]; p < 0.01) and severe night awakenings (B = -0.04 [-0.07;-0.02]; p < 0.01). To conclude, this study showed that some sleep problems can differentially predict earlier acceleration of deterioration in specific cognitive functions in older adults.

Abstract
BACKGROUND Sleep disorders are common health problems in the elderly. The effect of social isolation on sleep duration and sleep quality remains unclear in the Chinese community-dwelling elderly. AIM To explore the associations of social isolation with sleep duration and sleep quality in community-dwelling elderly people. METHODS A total of 7762 community-dwelling elderly people aged 60 years and older in Ningbo were recruited from June 2022 to August 2022. Data were collected using a questionnaire. Multivariate logistic regression was used to analyze the associations of social isolation with sleep duration and sleep quality. The KHB mediating effect model was used to test the mediating effect of depression and anxiety on these associations. RESULTS Among the participants, 2656 (34.2%) had abnormal sleep duration (< 6 hours or > 8 hours), 1115 (14.4%) had poor sleep quality, and 917 (11.8%) had social isolation. After adjusting for confounding factors, compared with the elderly without social isolation, the odds ratios (95%CI) of abnormal sleep duration and poor sleep quality in the elderly with social isolation were 1.49 (1.29-1.73) and 1.32 (1.09-1.59), respectively. Depression partially mediated the associations of social isolation with sleep duration and sleep quality, accounting for 5.68% and 9.87% of the mediating effect, respectively. CONCLUSION Social isolation was found to be associated with sleep duration and sleep quality in community-dwelling elderly people. More attention should be paid to social isolation in the elderly.

Abstract
Age-related changes in sleep have been associated with cognitive decline, yet causal pathways have not been identified. Evidence suggests reduced cardiovascular health may be a consequence of poor sleep and a precursor to cognitive decline. This observational cohort study used path analyses to determine whether cardiovascular disease risk mediated or moderated effects of sleep on yearly longitudinal change in cognition, estimated with linear growth models. Total sleep time (TST), sleep efficiency (SE), and relative spectral power of slow wave activity (SWA; 1-4 Hz) and slow oscillations (SO; 0.5-1 Hz), were measured with single-channel home EEG. Cardiovascular disease risk (CVR) was estimated as 10-year Framingham Risk Score 1-year post-sleep. Outcomes were yearly change in executive function (EF), episodic memory (EM), and processing speed (PS) over 2-5 years post-sleep. 342 participants (mean age 73.5 +/- 5.6 years, 51 % female) were included. Shorter TST was linearly associated with increased CVR across all models (βs = -0.18(0.058) - -0.19(0.059), ps< 0.002). TST was indirectly associated with EF and PS decline through CVR, such that associations between short TST and cognitive decline were partially due to higher CVR. All other mediating and moderating effects were nonsignificant after multiple comparisons. Indirect associations between short sleep duration and greater decline in executive function and processing speed were found through higher CVR, suggesting a potential mechanism by which sleep leads to cognitive decline. Findings support the prioritization of adequate sleep duration to preserve both cardiovascular and cognitive health in later life.

Abstract
Objective This study aims to investigate the relationship between sleep quality and cognitive impairment in middle-aged and older adults living in Western China. Methods Baseline data from the West China Health and Aging Trend (WCHAT) study were utilized, with enrollment of participants aged 50 years or older. Sleep quality was assessed using the Pittsburgh Sleep Quality Index (PSQI), where a PSQI score > 5 indicated poor sleep quality. The cognitive status was evaluated using the 10-item Short Portable Mental Status Questionnaire (SPMSQ). Multinomial logistic regression was applied to estimate odds ratios (ORs) and 95% confidence intervals (CIs). Results A total of 6,728 middle-aged and older adults dwelling in western China (age = 62.39 ± 8.925 years, 2,520 males and 4,208 females) were included in the analysis. The prevalence of mild cognitive impairment was 11.0%, with 4.3% having moderate to severe cognitive impairment. Poor sleep quality was found in 47.7% of participants. After adjusting for age, gender, education, marital status, and chronic disease, poor sleep quality (OR = 1.29, 95% CI: 1.1-1.52, p = 0.002) was associated with a higher risk of mild cognitive impairment. Among specific sleep dimensions, there is a significant association between high daytime dysfunction and mild cognitive impairment (adjusted OR = 1.96, 95% CI: 1.45-2.62) as well as moderate to severe cognitive impairment (adjusted OR = 3.15, 95% CI: 2.09-4.73) in the middle-aged and older adults residing in multi-ethnic areas of western China. Besides, a sleep duration of 6-7 h was associated with a reduced risk of mild cognitive impairment (OR = 0.80, 95% CI: 0.65-0.98) and moderate to severe cognitive impairment (OR = 0.71, 95% CI: 0.51-0.99). Stratified analysis showed that poor sleep quality-particularly daytime dysfunction-was significantly associated with moderate to severe cognitive impairment in Han (OR = 4.14, 95%CI: 1.65-10.23), Tibetan (OR = 3.45, 95% CI: 1.42-8.1), and Yi (OR = 4.04, 95%CI: 1.46-10.97) participants, but not in Uyghur and Qiang groups. Conclusion Sleep quality is significantly associated with cognitive impairment among middle-aged and older adults in Western China, particularly concerning the components of daytime dysfunction.

Abstract
BACKGROUND Sleep disorders are common among older adults and have a bidirectional impact on their emotional well-being. While some studies suggest that internet use may offer mental health benefits to this population, the relationship between internet use and sleep outcomes remains underexplored. OBJECTIVE This study investigates the association between internet use (including use frequency) and sleep quality and duration in middle-aged and older Chinese adults. METHODS A longitudinal analysis was conducted using the China Health and Retirement Longitudinal Study data from 2015 to 2018. Sleep quality was assessed using the sleep item from the Centre for Epidemiologic Studies Depression Scale, categorized as "good" (<1 day; reference), "fair" (1-4 days), or "poor" (5-7 days). Sleep duration was classified as short (<6 hours), medium (6-9 hours; reference), or long (>9 hours). Adjusted multinomial logistic regressions were used to examine the associations between internet use or frequency in 2015 and sleep quality or duration in 2018, controlling for age, sex, residence, diseases, smoking, drinking, and napping time and further exploring sex and age group variations. RESULTS The baseline analysis included 18,460 participants aged 45 years and older, with 1272 (6.9%) internet users, 8825 (48.1%) participants had fair or poor sleep, and 6750 (37.2%) participants had abnormal sleep duration. Internet users, particularly those who used it almost daily, were less likely to report poor sleep quality (relative risk [RR] 0.71, 95% CI 0.54-0.94) and longer sleep duration (RR 0.22, 95% CI 0.11-0.44) than nonusers. In the longitudinal analysis, baseline internet users had a significantly reduced risk of fair (RR 0.66, 95% CI 0.51-0.86) and poor sleep quality (RR 0.60, 95% CI 0.44-0.81), as well as short (RR 0.73, 95% CI 0.53-1.00) and long sleep duration (RR 0.39, 95% CI 0.21-0.72) during the follow-up period than nonusers. These associations remained significant for almost daily internet use (RR 0.32, 95% CI 0.15-0.69). Subgroup analyses by sex revealed a positive relationship between internet use and sleep quality, with a stronger effect in female (poor sleep: RR 0.57, 95% CI 0.36-0.89) than male (poor sleep: RR 0.61, 95% CI 0.40-0.92) participants. The effect on sleep duration was significant only in daily male users, showing a reduced risk of long sleep duration (RR 0.30, 95% CI 0.11-0.78). In the age subgroup analysis, most internet users were in the 45- to 59-year age group, with results consistent with the overall findings. CONCLUSIONS This study suggests that internet use is associated with a reduced risk of sleep problems in middle-aged and older adults. The findings indicate that moderate, regular internet engagement-such as daily use-may promote better sleep health in this population.

Abstract
Chronic pain conditions frequently coexist and share common genetic vulnerabilities. Despite evidence showing associations between pain and depression, the additive effect of co-occurring pain conditions on depression risk and the underlying mechanisms remain unclear. Leveraging data from 431,038 UK Biobank participants with 14-year follow-up, we found a significantly increased risk of depression incidence in individuals reporting pain, irrespective of body site or duration (acute or chronic), compared with pain-free individuals. The depression risk increased with the number of co-occurring pain sites. Mendelian randomization supported potential causal inference. We constructed a composite pain score by combining individual effects of acute or chronic pain conditions across eight body sites in a weighted manner. We found that depression risks increased monotonically in parallel with composite pain scores. Moreover, some inflammatory markers, including C-reactive protein, partially mediated the association between composite pain scores and depression risk. Considering the high prevalence of comorbid depression and pain, pain screening may help identify high-risk individuals for depression.

Abstract
Whether depression and poor sleep interact or have statistically independent associations with brain structure and its change over time is not known. Within a subset of UK Biobank participants with neuroimaging and subjective and/or objective sleep data (n = 28,351), we examined associations between lifetime depression and sleep disruption, and their interaction with structural neuroimaging measures, both cross-sectionally and longitudinally. Sleep variables were: self-reported insomnia and difficulty getting up; actigraphy-derived short sleep (<7 h); sustained inactivity bouts during daytime (SIBD); and sleep efficiency. Imaging measures were white matter microstructure, subcortical volumes, cortical thickness and surface area of 24 cortical regions of interest. Individuals with lifetime depression (self-reported, mental health questionnaire or health records) were contrasted with healthy controls. Interactions between depression and difficulty getting up for i) right nucleus accumbens volume and ii) mean diffusivity of forceps minor, reflected a larger negative association of poor sleep in the presence vs. absence of depression. Depression was associated with widespread reductions in white matter integrity. Depression, higher SIBD and difficulty getting up were individually associated with smaller cortical volumes and surface area, particularly in the frontal and parietal lobes. Many regions showed age-related decline, but this was not exacerbated by either depression or sleep disturbance. Overall, we identified widespread cross-sectional associations of both lifetime depression and sleep measures with brain structure. Findings were more consistent with additive rather than synergistic effects - although in some regions we observed greater magnitude of deleterious associations from poor sleep phenotypes in the presence of depression.

Abstract
BACKGROUND This study compares sleep duration and stroke risk between residents of China and the U.S. during and outside the COVID-19 pandemic, examining age as an interaction effect. METHODS This study analyzed data from the National Health and Nutrition Examination Survey (NHANES) and the China Health and Retirement Longitudinal Study (CHARLS). A total of 9131 American adults participated from 2017 to March 2020, and 7678 from August 2021 to August 2023. In China, 13,514 adults participated in 2018 and 9441 in 2020. Stroke incidence was assessed via survey responses, with "yes" indicating a history of stroke. Participants were categorized by age, and multivariable logistic regression and interaction analyses evaluated age effects, supported by subgroup and sensitivity analyses. Restricted cubic splines (RCS) examined nonlinear associations between sleep duration and stroke risk, while mediation analyses investigated the roles of hypertension, diabetes, and lipid abnormalities. RESULTS The study found that during the pandemic, in the multivariable-adjusted model, there was a "U-shaped" association between sleep duration and the prevalence of stroke (China: P for trend = 0.009, P non-linear = 0.0004; the United States: P for trend = 0.012, P non-linear = 0.0004). Similarly, in the multivariable-adjusted model, during the COVID-19 pandemic, compared with the non-pandemic period, long sleep duration (≥ 9 h) was potentially a risk factor for the prevalence of stroke among American adults (for those under 60 years old: odds ratio (OR) 95% confidence interval (CI)[1.836(1.138, 2.962)], P = 0.013; for those 60 years old and above: OR 95%CI[1.44(1.15, 1.436)], P = 0.044). In contrast, in China, compared with the pandemic period, the association between long sleep duration (≥ 9 h) and the prevalence of stroke was stronger during the non-pandemic period (for those 60 years old and above: OR 95%CI[1.342(1.132, 1.59)], P = 0.001), but no association was found among those under 60 years old. Mediation analysis indicated that in China, lipid abnormalities partially influenced the association between long sleep duration (≥ 9 h) and the prevalence of stroke. The mediation proportion was 8.39% in the overall population, and as high as 20.25% among the elderly aged 60 years and above. CONCLUSION During the COVID-19 pandemic, the association between prolonged sleep duration (≥ 9 h) and stroke risk among U.S. adults aged under 60 significantly increased, although this trend was less pronounced in China. These findings suggest that public health interventions should account for the varying impact of sleep duration across different populations.

Abstract
Genomic Copy Number variants (CNVs) increase risk for neurodevelopmental disorders (NDDs) and affect cognition, but their impact on sleep remains understudied despite the well-established link between sleep disturbances, NDDs, and cognition. We investigated the relationship between CNVs, sleep traits, cognitive ability, and executive function in 498,852 individuals from an unselected population in the UK Biobank. We replicated the U-shape relationship between measures of cognitive ability and sleep duration. The effects of CNVs on sleep duration were evident at the genome-wide level; CNV-burden analyses showed that overall, CNVs with an increasing number of intolerant genes were associated with increased or decreased sleep duration in a U-shape pattern (p < 2e-16), but did not increase risk of insomnia. Sleep duration only marginally mediated the robust association between CNVs and poorer cognitive performance, suggesting that sleep and cognitive phenotypes may result from pleiotropic effects of CNVs with minimal causal relationship.

Abstract
BACKGROUND Intrinsic capacity is a crucial indicator of adverse outcomes and has predictive value for diseases and mortality. The objective of this study is to assess the association between nighttime sleep duration and intrinsic capacity in older Chinese adults. METHODS Data were collected from the China Health and Retirement Longitudinal Study (CHARLS) spanning the years 2008-2020. Minimally sufficient adjustment was made for confounding factors identified through a Directed Acyclic Graphs (DAGs), and multivariate logistic regression analysis was conducted to determine the relationship between nighttime sleep duration and intrinsic capacity. Restricted cubic splines (RCS) were used to assess the nonlinear relationship between nighttime sleep duration and intrinsic capacity in this population. Finally, subgroup analysis and interaction effect analysis were performed. RESULTS Among the 4994 older adults, 3118 (62.4%) experienced a decline in intrinsic capacity. After adjusting for confounding factors, nighttime sleep duration was associated with an increased risk of intrinsic capacity decline (P < 0.001), exhibiting a nonlinear J-shaped pattern. CONCLUSIONS For Chinese older adults, increased nighttime sleep duration shows a nonlinear, J-shaped dose-response relationship with the risk of intrinsic capacity decline. Our study provides important insights into the intrinsic capacity and health self-management of older adults.

Abstract
Background With the acceleration of population aging, cognitive impairment and depression have become serious public health challenges in countries around the world. The influencing factors of cognitive trajectory, depression trajectory, and dual trajectories in middle-aged and elderly adults have not been fully studied. Methods This study used data from the China Health and Retirement Longitudinal Study database spanning from 2011-2018. Group-based trajectory modelling and group-based dual trajectory modelling were employed to examine different trajectories. Restricted cubic spline and multivariate logistic regression analysis were used to elucidate the relationship between sleep duration and grip strength with these different trajectories. Mediation analysis was conducted to explore the mediating roles of sleep duration and grip strength in the activities of daily living (ADLs) and their impact on these trajectories. Results Trajectory analysis identified two longitudinal patterns of cognitive function and depression scores: low and high cognitive group, low and high depression group, respectively, and two states of the dual trajectories of cognition and depression: the stable state group and the state decline group. Sleep duration and grip strength were associated with the cognitive trajectory, depression trajectory and dual trajectories. Sleep duration has an inverted U-shaped relationship with cognitive trajectory. Grip strength was nonlinearly associated with the above trajectories. The mediation effects of sleep duration in the association between ADLs and cognitive, depression and dual trajectories were 3.14, 6.14, and 2.70%. While the mediation effects of grip strength were 7.21, 1.67 and 6.24%, respectively (P < 0.05). Conclusions Sleep duration and grip strength were not only associated with cognitive, depression, and dual trajectories, but also partially mediate the relationship between ADLs and these trajectories. This study will provide a basis for how to intervene in the cognitive and mental health of middle-aged and elderly adults.

Abstract
In the current study we introduce statistical methods based on trigonometry, to infer the shape of a (non)-linear bivariate genetic relationship. We do this based on a series of piecemeal GWASs of segments of a target (continuous) trait distribution, and the genetic correlations between those GWASs and a second trait. Simulations confirm that we are able to retrieve the shape of the relationship given certain assumptions about the nature of the relationship between the traits. We applied the method to the genetic relationship between BMI, sleep duration, and height, and psychiatric disorders (ADHD, anorexia nervosa, and depression) using data from approximately 450K individuals from UK Biobank. In the relationship between BMI and psychiatric traits, we found that the expected value of depression is a nonlinear function of BMI i.e. there is a nonlinear genetic relationship between both traits. We observed similar findings for the genetic relationship between BMI and anorexia, sleep duration and depression, and sleep duration and ADHD. We observed no underlying nonlinearity in the genetic relationship between height and psychiatric traits. Using a novel statistical approach, we show that nonlinear genetic relationships between traits are detectable and genetic associations as quantified using global estimators like genetic correlations are not informative about underlying complexities in these relationships. Our findings challenge assumptions of linearity in genetic epidemiology and suggest that bivariate genetic associations are not uniform across the phenotypic spectrum, which may have implications for the development of targeted interventions.

Abstract
OBJECTIVES This study aimed to explore the causal association between sleep duration and depression in adolescents and young adults. METHODS We conducted a systematic review and Mendelian randomization (MR) to research the causal relationship between short sleep duration and adolescent depression risk from an observational and genetic perspective. In the systematic review, we searched observational studies from the PubMed, Embase, and Cochrane Library databases. In the MR analysis part, we screened Single Nucleotide Polymorphism (SNP) significantly relative to short sleep and conforming MR concept to investigate the genetic causality. RESULTS All research evidence shows that adolescents who sleep <6 h have the highest rates of depression. According to the MR result, short sleep duration significantly affected the depression risk (odds ratio, 1.034; 95% confidence interval, 1.012-1.058, P = .003). Sleep duration of 7-8 h has the lowest depression incidence. Insufficient sleep (≤6 h) and excessive sleep (≥8 h) also elevates adolescent depression risk. Genetic evidence shows that short sleep duration (<6 h) has significant causal effects on depression risk. CONCLUSIONS Sleep duration was causally associated with depression in adolescents and young adults. Sleep duration of <6 h or >8 h daily increases the depression risk in adolescents and young adults.

Abstract
While animals across species typically experience suppressed consciousness and an increased arousal threshold during sleep, the responsiveness to specific sensory inputs persists. Previous studies have demonstrated that rhythmic and continuous vibration can enhance sleep in both animals and humans. However, the neural circuits underlying vibration-induced sleep (VIS) and its potential therapeutic benefits on neuropathological processes in disease models remain unclear. Here, it is shown that intermittent vibration, such as cycles of 30 s on followed by 30 s off, is more effective in inducing sleep compared to continuous vibration. A clear evidence is further provided that allatostatin A (AstA)-GABA signaling mediates short-term intermittent vibration-induced sleep (iVIS) by inhibiting octopaminergic arousal neurons through activating GABAA receptors. The existence of iVIS in mice is corroborated, implicating the GABAergic system in this process. Finally, intermittent vibration not only enhances sleep but also reduces amyloid-β (Aβ) deposition and reverses memory defects in Alzheimer's disease models. In conclusion, the study defines a central neural circuit involved in mediating short-term iVIS and the potential implications of vibration in treating sleep-related brain disorders.

Abstract
BACKGROUND This study aimed to investigate age-specific alterations in functional connectivity after sleep deprivation (SD) and decode brain functional changes from neurocognitive and transcriptomic perspectives. METHODS Here, we examined changes in global and regional graph measures, particularly regional network strength (RNS), in 41 young participants and 36 older participants with normal sleep and after 3 h of SD. Additionally, by utilizing cognitive probabilistic maps from Neurosynth and gene expression data from the Allen Human Brain Atlas, we applied partial least-squares regression analysis to identify the neurocognitive and transcriptional correlates of these RNS changes. RESULTS After SD, older participants exhibited decreased RNS in the default mode network (DMN) and dorsal attention network, with increased RNS in the visual network. Young participants also showed decreased RNS in the DMN, notably in the left inferior parietal lobe, left dorsolateral prefrontal cortex, and left posterior cingulate cortex. In young participants, SD-induced RNS changes significantly correlated with cognitive processes such as "attention," "cognitive control," and "working memory," while in older participants, they correlated with "learning," "focus," and "decision." Gene-category enrichment analysis indicated that specific genes related to signal transduction, ion channels, and immune signaling might influence SD pathophysiology by affecting functional connectivity in young participants. CONCLUSIONS This study elucidates shared and age-specific brain functional network alterations associated with SD, providing a neurocognitive and molecular basis for understanding the underlying pathophysiology.

Abstract
The Centers for Disease Control and Prevention declared sleep-related problems to be a public health epidemic. With the advent of biometric sleep tracking technology taking the sleep lab into the field, the study of human sleep is now global, and these new datasets show contrasting findings. Previous reports suggest sleep in small-scale, non-industrial societies to be short and fragmented yet characterized by greater circadian rhythmicity. However, the role of circadian rhythm indicators in understanding global variations in human sleep patterns remains unclear. We examine population-level sleep studies (n = 54) using polysomnography and actigraphy to test the sleep restriction epidemic hypothesis, which posits that labour demands and technological disruption in large-scale, industrial societies have reduced sleep duration. We used an actigraphy-generated circadian function index from both non-industrial and industrial societies (n = 866) to test the circadian mismatch hypothesis, which suggests that poor chronohygiene in regulated environments misaligns circadian rhythms in industrial societies. In rejection of the sleep restriction epidemic hypothesis, our results show that industrial societies experience the longest, most efficient sleep, whereas in support of the circadian mismatch hypothesis, sleepers in non-industrial societies are characterized by the greatest circadian function.

Abstract
BACKGROUND Epilepsy, as a chronic noncommunicable disease with recurrent seizures, may be a marker of deterioration or alteration in other underlying neurological diseases. This study aimed to investigate the relationship of epilepsy with brain function, other common brain disorders, and their underlying mechanisms. METHODS The study was based on clinical diagnostic and test data from 426,527 participants in the UK Biobank, of whom 3,251 were diagnosed with epilepsy at baseline. Multiple linear and Cox regression models were used to explore the association between epilepsy, brain function, and other brain disorders. RESULTS This study demonstrated consistent deleterious effects of epilepsy on cognitive and motor function and mental health. The risk of neurological diseases and psychiatric disorders was significantly elevated in the epilepsy population during the 17-year follow-up period, according to the longitudinal analysis. We also identified several brain regions associated with epilepsy, including the pallidum, hippocampus, and precentral regions. Mediation analyses revealed mediating effects of peripheral markers and proteins (e.g., GGT, HDL, ACE2, and GDF15), suggesting that liver function and lipid metabolism may be involved in the development of other brain disorders in individuals with epilepsy. CONCLUSIONS Our study provides robust evidence of the association between epilepsy and poor brain health, underscoring the importance of early intervention for epilepsy.

Abstract
Background: The occurrence of sleep disturbances in amyotrophic lateral sclerosis (ALS) patients is widely reported. However, there is still a lack of reliable evidence of a relationship between sleep disturbances and the risk of developing ALS. The aim of this study was to prospectively investigate the longitudinal associations between sleep traits and the risk of incident ALS. Methods: We included information from 409,045 individuals from the prospective cohort of the UK Biobank. Sleep traits at baseline were measured using a standardized questionnaire. All sleep traits were analyzed in relation to the subsequent incidence of ALS using Cox proportional hazards models. Results: Multivariate analysis showed that 6-7 h of sleep was related to the lowest risk for ALS. A long sleep duration (≥8 h) was associated with an increased risk of ALS incidence (HR: 1.31, 95% CI: 1.07-1.61; p = 0.009). A short sleep duration (<6 h) was associated with an increased risk of ALS incidence (HR: 1.91, 95% CI: 1.10-3.30, p = 0.021) in females. In participants aged ≥65 years, eveningness was associated with increased ALS risk (HR: 1.32, 95% CI: 1.08-1.61; p = 0.006). Conclusion: Our results hint at a sleep duration that is too short or too long, and certain chronotypes might be related to the risk of developing ALS. Despite the limitations imposed by the study design and the subjectivity of sleep information, our findings suggest that sleep disturbances may influence the risk of developing ALS.

Abstract
Recent genetic studies have linked numerous loci to psychiatric disorders. However, the biological pathways that connect these genetic associations to psychiatric disorders' specific pathophysiological processes are largely unclear. Endophenotypes, first defined over five decades ago, are heritable traits, independent of disease state that are associated with a disease, encompassing a broad range of neurophysiological, biochemical, endocrinological, neuroanatomical, cognitive, and neuropsychological characteristics. Considering the advancements in genetics and genomics over recent decades, we propose a revised definition of endophenotypes as 'genetically influenced phenotypes linked to disease or treatment characteristics and their related events.' We also updated endophenotype criteria to include (1) reliable measurement, (2) association with the disease or its related events, and (3) genetic mediation. 'Genetic mediation' is introduced to differentiate between causality and pleiotropic effects and allows non-linear relationships. Furthermore, this updated Endophenotype 2.0 framework expands to encompass genetically regulated responses to disease-related factors, including environmental risks, illness progression, treatment responses, and resilience phenotypes, which may be state-dependent. This broadened definition paves the way for developing new endophenotypes crucial for genetic analyses in psychiatric disorders. Integrating genetics, genomics, and diverse endophenotypes into multi-dimensional mechanistic models is vital for advancing our understanding of psychiatric disorders. Crucially, elucidating the biological underpinnings of endophenotypes will enhance our grasp of psychiatric genetics, thereby improving disease risk prediction and treatment approaches.

Abstract
The pivotal role of sleep has led to extensive research endeavors aimed at automatic sleep stage classification. However, existing methods perform poorly when classifying small groups or individuals, and these results are often considered outliers in terms of overall performance. These outliers may introduce bias during model training, adversely affecting feature selection and diminishing model performance. To address the above issues, this paper proposes an ensemble-based sequential convolutional neural network (E-SCNN) that incorporates a clustering module and neural networks. E-SCNN effectively ensembles machine learning and deep learning techniques to minimize outliers, thereby enhancing model robustness at the individual level. Specifically, the clustering module categorizes individuals based on similarities in feature distribution and assigns personalized weights accordingly. Subsequently, by combining these tailored weights with the robust feature extraction capabilities of convolutional neural networks, the model generates more accurate sleep stage classifications. The proposed model was verified on two public datasets, and experimental results demonstrate that the proposed method obtains overall accuracies of 84.8% on the Sleep-EDF Expanded dataset and 85.5% on the MASS dataset. E-SCNN can alleviate the outlier problem, which is important for improving sleep quality monitoring for individuals.

Abstract
Background/Objectives: A significant number of patients experience prolonged symptoms following COVID-19 in particular cases of fatigue. Yet, pathomechanisms of COVID-19-related fatigue remain unclear. Methods: This study investigated patients after confirmed SARS-CoV-2 infection (PC) with relevant fatigue according to the Fatigue Assessment Scale (≥22 points) to rule out malnutrition as a driving factor for fatigue and to evaluate daily activity and sleep characteristics. Dietary behavior was recorded through food diaries and physical activity by self-reported (questionnaires) and objective (activity tracker) outcomes. Data were collected over a 7-day period and compared with a healthy control group (HC). A subgroup analysis of patients with fatigue and severe fatigue, as well as a sex-specific analysis, were included. Results: No significant differences in dietary intake were observed, but an indication toward a healthier Mediterranean diet in PC patients with a median Mediterranean Diet Score of 4 (IQR 3, 5) in HC vs. 5 (IQR 3, 6) in PC (p = 0.24). There were also no differences in physical activity, either by objective or subjective measures. However, the median sleep duration was 49 min longer in PC patients (p = 0.003). Conclusions: In conclusion, malnutrition did not significantly contribute to fatigue, yet patients with COVID-19-related fatigue showed increased sleep duration. As sleep characteristics play a crucial role in mental and physical wellbeing, the association of sleep, physical activity, and fatigue should be evaluated in further studies.

Abstract
OBJECTIVES This study aimed to investigate the association between sleep disorders and the prevalence of taste dysfunction and the mediation effect of oral microbe in adults over 40 years. MATERIALS AND METHODS Cross-sectional data were utilized from the National Health and Nutrition Examination Survey (2011-2014). Regression models were employed, adjusting for demographic variables and covariates. Subgroup analyses were conducted based on age, sex, ethnicity, and education level. Multiplicative interactions were assessed through likelihood ratio tests. Additionally, the impact of sleep disturbance on the alpha diversity of the oral microbiome was examined using the rank-sum test (significance threshold: p < 0.05). Mediation analysis based on oral microbiota was conducted. RESULTS The analysis included 4869 participants. After adjusting for adjusting for demographic variables and covariates, individuals with sleep disorders exhibited a 36% increased risk of taste dysfunctions compared to those without sleep disorders (OR: 1.36, 95% CI: 1.00-1.84, p = 0.05). Interaction analyses indicated no significant differences between sleep disorders and taste dysfunctions concerning sex, educational level, and age across various models (Crude Model, Model 1, Model 2, and Model 3; p for interaction > 0.05). Furthermore, compared with the non-sleep disorder group, patients with sleep disorders demonstrated decreased numbers of OTUs, Shannon-Wiener indices, and Faith's phylogenetic diversity indices in the oral microbiota (p < 0.05). However, the mediation analysis failed to reveal an indirect effect of oral microbiome on taste dysfunction (p > 0.05.) CONCLUSION: Sleep disorders independently correlate with a higher risk of taste dysfunctions, potentially associated with alterations in oral flora.

Abstract
Aging is the result of a complex interplay of physical, environmental, and social factors, leading to an increased prevalence of chronic age-related diseases that burden health and social care systems. As the global population ages, it is crucial to understand the aged immune system, which undergoes declines in both innate and adaptive immunity. This immune decline exacerbates the aging process, creating a feedback loop that accelerates the onset of diseases, including infectious diseases, autoimmune disorders, and cancer. Intervention strategies, including dietary adjustments, pharmacological treatments, and immunomodulatory therapies, represent promising approaches to counteract immunosenescence. These interventions aim to enhance immune function by improving the activity and interactions of aging-affected immune cells, or by modulating inflammatory responses through the suppression of excessive cytokine secretion and inflammatory pathway activation. Such strategies have the potential to restore immune homeostasis and mitigate age-related inflammation, thus reducing the risk of chronic diseases linked to aging. In summary, this review provides insights into the effects and underlying mechanisms of immunosenescence, as well as its potential interventions, with particular emphasis on the relationship between aging, immunity, and nutritional factors.

Abstract
OBJECTIVES Intrinsic capacity (IC), a multidimensional construct encompassing mental and physical capacities, has been established in the aging framework by the World Health Organization. However, the detailed relationship between IC and Chinese sleep patterns (nighttime sleep and post-lunch naps) remains inadequately elucidated. METHODS Participants in this study were individuals aged ≥45 years residing in China, included in the China Health and Retirement Longitudinal Study (CHARLS). We analyzed 4 years of CHARLS data from the first wave (May 2011-March 2012) to the second wave (July 2015-January 2016). Data from these waves were utilized for longitudinal analysis. Self-reported data included nighttime sleep and nap duration, along with other baseline characteristics. The IC evaluation involved physical examinations and blood tests. Initially, linear regression was used to assess the relationship between total sleep duration, nighttime sleep duration, nap duration, and IC change between the two waves that were determined by marginal effects (ME) and their corresponding 95% confidence intervals (CIs). Regression splines were employed to explore potential nonlinear associations. Subgroup and sensitivity analyses were conducted to investigate the heterogeneity of IC change under specific conditions and the robustness of our results. Mediation analysis was performed to identify potential factors mediating the relationship between sleep patterns and IC change. RESULTS Both excessive (>10 h) (total, ME: -1.12; 95% CI: -1.61, -0.64; nighttime, ME: -1.44; 95% CI: -2.29, -0.59) and insufficient (<6 h) sleep duration (total, ME: -0.43; 95% CI: -0.68, -0.18; nighttime, ME: -0.50; 95% CI: -0.73, -0.27) negatively impacted IC change. Moderate naps (≤60 min) mitigated the decline in IC change (ME: 0.28; 95% CI: 0.07, 0.49). IC values decreased at the slowest rate when nap time constituted one-seventh of total sleep time. The onset of dyslipidemia partially mediated the association between naps (≤60 min) and IC change (P = 0.02). CONCLUSIONS These findings suggest that maintaining a healthy sleep pattern of 6-8 h of nighttime or total sleep, along with a post-lunch nap of ≤60 min, helps preserve optimal IC or delay its decline. This is particularly beneficial for cognitive, psychological, and locomotion performance among middle-aged and older adults.

Abstract
Aging is associated with declines in cognition and brain structural integrity. However, there is equivocality over (1) the specificity of affected domains in different people, (2) the location of associated patterns of brain structural deterioration, and (3) the sociodemographic factors contributing to 'unhealthy' cognition. We aimed to identify cognitive profiles displayed by older adults and determine brain and sociodemographic features potentially shaping these profiles. A sample of Southeast-Asian older adults (N = 386) participated in a multi-session study comprising cognitive testing, neuroimaging, and a structured interview. We used computational models to extract latent mechanisms underlying cognitive flexibility and response inhibition. Data-driven methods were used to construct cognitive profiles based on standard performance measures and model parameters. We also investigated grey matter volume and machine-learning derived 'brain-ages'. A profile associated with poor set-shifting and rigid focusing was associated with widespread grey matter reduction in cognitive control regions. A slow responding profile was associated with advanced brain-age. Both profiles were correlated with poor socioeconomic standing and cognitive reserve. We found that the impact of sociodemographic factors on cognitive profiles was partially mediated by total grey and white matter, and dorsolateral prefrontal and cerebellar volumes. This study furthers understanding of how distinct aging profiles of cognitive impairment uniquely correspond to specific vs. global brain deterioration and the significance of socioeconomic factors in informing cognitive performance in older age.

Abstract
BACKGROUND Poor sleep quality is increasingly considered to be an underlying cause of cerebrovascular diseases. This is a slowly progressing condition that gradually leads to vascular cognitive impairment and stroke during ageing. At present, randomized clinical trials examining the non-pharmacological therapies in the management of this comorbidity are very limited. Transcranial current stimulation (tCS) is a non-invasive technology for promoting cognitive function and treating brain disorders. As advanced modalities of tCS, transcranial random noise stimulation (tRNS) and transcranial alternating current stimulation (tACS), could deliver frequency-specific waveforms of currents that can modulate brain activities in a more specific manner. METHODS AND DESIGN Chinese individuals between the ages of 60 and 90 years, who are right-handed and have mild vascular cognitive impairment (VCI) with sleep disturbances, will participate in a randomized study. They will undergo a 2-week intervention period where they will be randomly assigned to one of three groups: high-frequency (hf)-tRNS, 40 Hz tACS, or sham tCS. Each group will consist of 15 participants. Before the intervention, high-resolution magnetic resonance imaging (MRI) data will be used to create a computational head model for each participant. This will help identify the treatment target of left inferior parietal lobe (IPL). Throughout the study, comprehensive assessments will be conducted at multiple time points, including baseline, 2nd week, 6th week, and 12th week. These assessments will evaluate various factors such as sleep quality, domain-specific cognitive performance, and actigraphic records. In addition, the participants' adherence to the program and any potential adverse effects will be closely monitored throughout the duration of the intervention. CONCLUSIONS The primary objective of this study is to examine the safety, feasibility, and effectiveness of hf-tRNS and 40 Hz tACS interventions targeting left IPL in individuals with mild vascular cognitive impairment (VCI) who experience sleep disturbances and cognitive dysfunction. Additionally, the study seeks to evaluate the program's adherence, tolerability, and any potential adverse effects associated with frequency-specific transcranial current stimulation (tCS). The findings from this research will contribute to a deeper understanding of the intricate relationship between oscillation, sleep, and cognition. Furthermore, the results will provide valuable insights to guide future investigations in the field of sleep medicine and neurodegenerative diseases. Abstract
BACKGROUND The prospective association between sleep duration and the development of late-life depressive symptomology is unclear. AIMS To investigate sleep duration from midlife to late life in relation to risk of depressive symptoms in late life. METHOD A total of 14 361 participants from the Singapore Chinese Health Study were included in the present study. Daily sleep duration was self-reported at baseline (mean age of 52.4 years; 1993-98), follow-up 2 (mean age of 65.2 years; 2006-10) and follow-up 3 (mean age of 72.5 years; 2014-16) interviews. Depressive symptoms were evaluated using the Geriatric Depression Scale at follow-up 3 interviews. Modified Poisson regression models were performed to estimate relative risks and 95% confidence intervals of late-life depressive symptoms in relation to sleep duration at baseline and the two follow-up interviews. RESULTS Compared with sleeping 7 h per day, a short sleep duration of ≤5 h per day at baseline (i.e. midlife) was related to a higher risk of depressive symptoms (relative risk 1.10, 95% CI 1.06-1.15), and this risk was not affected by subsequent prolongation of sleep. Conversely, a long sleep duration of ≥9 h per day at baseline was not related to risk of depressive symptoms. At follow-up 3 (i.e. late life), both short sleep (relative risk 1.20, 95% CI 1.16-1.25) and long sleep (relative risk 1.12, 95% CI 1.07-1.18) duration were cross-sectionally associated with depressive symptoms. CONCLUSION Short sleep duration in midlife, regardless of subsequent prolongation, is associated with an increased risk of depression in late life. Contrariwise, both short and long sleep duration in late life co-occur with depressive symptoms.

Abstract
Accumulating evidence indicates that individuals with chronic kidney disease (CKD) are at an increased risk of experiencing depressive disorders, which may accelerate its progression. However, the relationship between the triglyceride-glucose (TyG) index and depression in CKD individuals remains unclear. Therefore, this cross-sectional study aimed to assess whether such a relationship exists. To this end, the CKD cohort of the National Health and Nutrition Examination Survey from 2005 to 2020 was analyzed using multivariable logistic regression analyses and a generalized additive approach. A recursive algorithm was employed to pinpoint the turning point, constructing a dual-segment linear regression model. The study included 10,563 participants. After controlling for all variables, the odds ratios and 95% confidence intervals indicated a 1.24 (range, 1.09-1.42) relationship between the TyG index and depression in the CKD cohort. The findings underscored an asymmetrical association, with a pivotal value at a TyG index 9.29. Above this threshold, the adjusted odds ratio (95% confidence interval) was 1.10 (range, 0.93-1.31). This relationship was significant among the obese subgroups. The study results highlight the complex relationship between the TyG index and depression among American adults with CKD.

Abstract
The Econeurobiology of the brain describes the environment in which an individual's brain develops. This paper explores the complex neural mechanisms that support and evaluate enrichment at various stages of development, providing an overview of how they contribute to plasticity and enhancement of both achievement and health. It explores the deep benefits of enrichment and contrasts them with the negative effects of trauma and stress on brain development. In addition, the paper strongly emphasizes the integration of Gardner's intelligence types into the school curriculum environment. It emphasizes the importance of linking various intelligence traits to educational strategies to ensure a holistic approach to cognitive development. In the field of Econeurobiology, this work explains the central role of the environment in shaping the development of the brain. It examines brain connections and plasticity and reveals the impact of certain environmental factors on brain development in early and mid-childhood. In particular, the six key factors highlighted are an environment of support, nutrition, physical activity, music, sleep, and cognitive strategies, highlighting their potential to improve cognitive abilities, memory, learning, self-regulation, and social and emotional development. This paper also investigates the social determinants of health and education in the context of Econeurobiology. It emphasizes the transformative power of education in society, especially in vulnerable communities facing global challenges in accessing quality education.

Abstract
Background This study aims to explore the association between sleep duration and the prevalence of chronic musculoskeletal pain (CMP). Methods A cross-sectional study was conducted using data from the National Health and Nutrition Examination Survey (NHANES) 2009-2010, which involved multiple centers across the United States. The study included 3,904 adults selected based on age and complete data availability. Demographic variables such as gender, age, race, and socioeconomic status (represented by the poverty-to-income ratio) were considered. Results Of the participants, 1,595 reported less than 7 h of sleep, 2,046 reported 7-8 h, and 263 reported more than 9 h of sleep. Short sleep duration was associated with higher odds of CMP (OR, 1.611, 95% CI: 1.224-2.120, p = 0.005). Long sleep duration also showed a higher prevalence (OR, 1.751; 95% CI, 0.923 to 3.321; p = 0.059), although this result was not statistically significant. A U-shaped relationship emerged (Effective degree of freedom (EDF) = 3.32, p < 0.001), indicating that 7 h of sleep was associated with the lowest odds of CMP. In individuals with sleep durations less than 7 h, each hour increment correlated with 22.8% reduced odds of CMP (OR, 0.772; 95% CI, 0.717-0.833; p = 0.002). Beyond 7 h, each hour increment was associated with 38.9% increased odds of CMP (OR, 1.389; 95% CI, 1.103-1.749; p = 0.049). Conclusion The findings suggest that both insufficient and excessive sleep durations are linked to a higher prevalence of CMP, highlighting the importance of optimal sleep duration for musculoskeletal health.

Abstract
Although it is generally recognized that sleep quality, depressive symptoms, and cognitive functions are related respectively, the main ambiguity comes from difficulties in determining their cause-effect relationships. The present study aimed to explore the longitudinal causation relationships among sleep quality, depressive symptoms, and cognitive functions in older people with mild cognitive impairment (MCI). A total of 134 patients from 24 communities in Ningbo City, Zhejiang Province, China with MCI were interviewed at baseline, while 124 of them were re-interviewed 2 months later, and 122 were re-interviewed 4 months later. The Patient Health Questionnaire-9, the Pittsburgh Sleep Quality Index and the Montreal Cognitive Assessment Scale were assessed in the interview. Cross-lagged models were tested to disentangle the relationships among sleep quality, depressive symptoms, and cognitive functions using structural equation modeling with latent variables on the four-mouth longitudinal data. The correlation coefficients between sleep quality and depressive symptoms were significant showing the stability across time points of assessment, while the correlation coefficient of cognitive function was not significant (r = 0.159, p > 0.05). The results of index of model fit indicated that the cross-lagged model was acceptable (CFI = 0.934, TLI = 0.899, RMSEA = 0.075, χ2/df = 1.684). The results of cross-lagged model analysis supported the complete mediating role of depressive symptoms in the association between sleep quality and cognitive functions, where worse sleep quality may lead to more severe depressive symptoms, which in turn leads to more severe cognitive decline. In Conclusion, sleep quality is significantly correlated with cognitive functions in patients with mild cognitive impairment, which association is fully mediated by depressive symptoms. Approaches addressing sleep quality and depressive symptoms are recommended and hold promise for the management of mild cognitive impairment.

Abstract
BACKGROUND High-density lipoprotein cholesterol (HDL-C) is widely recognized for its protective effects against cognitive decline. However, recent studies have presented conflicting results, with some suggesting no significant cognitive benefits or even an increased risk of dementia associated with high HDL-C levels. For those who suffer from depression, the cognitive benefits of HDL-C may be diminished or reversed. The purpose of this study is to investigate the associations between HDL-C, cognitive ability, and depressive symptoms in middle-aged and older Chinese adults. METHODS The datasets utilized were sourced from the China Health and Retirement Longitudinal Study (CHARLS) for the years 2011 and 2015, comprising 4,302 participants. Cross-lagged models were employed to explore the temporal sequence between cognitive performance and HDL-C levels, and to examine the interplay among depression, cognition, and HDL-C. Confounding factors such as sociodemographic characteristics, sleep conditions, and history of chronic diseases were controlled for. RESULTS The analysis revealed unidirectional effects of baseline impaired cognition and greater severity of depression on increased HDL-C levels at follow-up (β = - 0.036 and β = 0.028, respectively, P < 0.05). However, higher baseline HDL-C levels did not significantly predict cognitive performance or depression 4 years later (β = - 0.008 and β = 0.023, respectively, P > 0.05). Depressive symptoms and cognition were found to have a significant bidirectional association (β = - 0.026 and β = - 0.053, respectively, P < 0.05). CONCLUSIONS Cognitive impairment and depression are associated with higher HDL-C levels, whereas higher HDL-C levels do not appear to protect against cognitive decline or depressive symptoms. These findings underscore the importance of preserving cognitive and mental health, which may lower the likelihood of cardiovascular disease and dementia. Future studies should validate these findings and develop targeted interventions tailored to specific populations.

Abstract
Poor sleep health is associated with increased all-cause mortality and incidence of many chronic conditions. Previous studies have relied on cross-sectional and self-reported survey data or polysomnograms, which have limitations with respect to data granularity, sample size and longitudinal information. Here, using objectively measured, longitudinal sleep data from commercial wearable devices linked to electronic health record data from the All of Us Research Program, we show that sleep patterns, including sleep stages, duration and regularity, are associated with chronic disease incidence. Of the 6,785 participants included in this study, 71% were female, 84% self-identified as white and 71% had a college degree; the median age was 50.2 years (interquartile range = 35.7, 61.5) and the median sleep monitoring period was 4.5 years (2.5, 6.5). We found that rapid eye movement sleep and deep sleep were inversely associated with the odds of incident atrial fibrillation and that increased sleep irregularity was associated with increased odds of incident obesity, hyperlipidemia, hypertension, major depressive disorder and generalized anxiety disorder. Moreover, J-shaped associations were observed between average daily sleep duration and hypertension, major depressive disorder and generalized anxiety disorder. These findings show that sleep stages, duration and regularity are all important factors associated with chronic disease development and may inform evidence-based recommendations on healthy sleeping habits.

Abstract
OBJECTIVE To explore the latent profile of depression in older adults raising grandchildren in China, and to analyze the differences in the characteristics of the different profiles of the population and the factors influencing them. METHODS This study utilized the 2018 China Health and Retirement Longitudinal Study (CHARLS). Through a cross-sectional study, the latent profile analysis was used to analyze the potential classification of depression among older adults raising grandchildren, and using ordered multi-categorical logistic regression analyses to assess the effects of each factor on their different classifications. RESULTS The 1271 older adults raising grandchildren with depression symptoms were divided into three categories: low-level depression (55.4%), moderate-level depression (31.2%), and high-level depression (13.4%). Ordered multi-categorical Logistic results showed: Gender, marital status, pension insurance, physical health status, life satisfaction, and IADL were predictors of latent profile classification of depression symptoms in older adults raising grandchildren (P < 0.05). CONCLUSIONS In the future of primary care, it will be more meaningful to provide targeted interventions for different subgroups of depression in older adults raising grandchildren.

Abstract
Consuming probiotic products is a solution that people are willing to choose to augment health. As a global health hazard, sleep deprivation (SD) can cause both physical and mental diseases. The present study investigated the protective effects of Lacticaseibacillus rhamnosus GG (LGG), a widely used probiotic, on a SD mouse model. Here, it has been shown that SD induced intestinal damage in mice, while LGG supplementation attenuated disruption of the intestinal barrier and enhanced the antioxidant capacity. Microbiome analysis revealed that SD caused dysbiosis in the gut microbiota, characterized by increased levels of Clostridium XlVa, Alistipes, and Desulfovibrio, as well as decreased levels of Ruminococcus, which were partially ameliorated by LGG. Moreover, SD resulted in elevated pro-inflammatory cytokine concentrations in both the intestine and the brain, while LGG provided protection in both organs. LGG supplementation significantly improved locomotor activity in SD mice. Although heat-killed LGG showed some protective effects in SD mice, its overall efficacy was inferior to that of live LGG. In terms of mechanism, it was found that AG1478, an inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase, could diminish the protective effects of LGG. In conclusion, LGG demonstrated the ability to alleviate SD-induced intestinal barrier dysfunction through EGFR activation and alleviate neuroinflammation.

Abstract
Study Objectives Evidence suggests that poor sleep impacts cognition, brain health, and dementia risk but the nature of the association is poorly understood. This study examined how self-reported sleep duration, napping, and subjective depression symptoms are associated with the brain-cognition relationship in older adults, using sulcal width as a measure of relative brain health. Methods A canonical partial least squares analysis was used to obtain two composite variables that relate cognition and sulcal width in a cross-sectional study of 137 adults aged 46-72. We used a combination of ANCOVA and path analyses to test the associations of self-reported sleep duration, napping, and subjective depression symptoms with the brain-cognition relationship. Results We observed a significant main effect of sleep duration on sulcal width, with participants reporting 7 hours showing narrower sulci than other durations. This effect remained significant after including subjective depression as a covariate, which also had a significant main effect on sulcal width in the model. There was no significant effect of napping on sulcal width. In path analyses where the effects of age, self-reported sleep duration and depression symptoms were investigated together, sulcal width mediated the relationship between age and cognition. We also observed a significant indirect effect of sulci width in the subjective depression-cognition relationship. Conclusions Findings suggest that self-reported sleep duration and subjective depression may each be independently associated with brain morphology, which is related to cognitive functions. Results could help inform clinical trials and related intervention studies that aim at delaying cognitive decline in adults at risk of developing dementia.

Abstract
Sleep is a fundamental state for maintaining the organism homeostasis. Disruptions in sleep patterns predispose to the appearance of memory impairments and mental disorders, including depression. Recent pre-clinical studies have highlighted the antidepressant-like properties of the synthetic compound 2-phenyl-3-(phenylselanyl)benzofuran (SeBZF1). To further investigate the neuromodulatory effects of SeBZF1, this study aimed to assess its therapeutic efficacy in ameliorating neurobehavioral impairments induced by sleep deprivation (SD) in mice. For this purpose, a method known as multiple platforms over water was used to induce rapid eye movement (REM) SD. Two hours after acute SD (24 h), male Swiss mice received a single treatment of SeBZF1 (5 mg/kg, intragastric route) or fluoxetine (a positive control, 20 mg/kg, intraperitoneal route). Subsequently, behavioral tests were conducted to assess spontaneous motor function (open-field test), depressive-like behavior (tail suspension test), and memory deficits (Y-maze test). Brain structures were utilized to evaluate oxidative stress markers, monoamine oxidase (MAO) and acetylcholinesterase (AChE) activities. Our findings revealed that SD animals displayed depressive-like behavior and memory impairments, which were reverted by SeBZF1 and fluoxetine treatments. SeBZF1 also reverted the increase in lipoperoxidation levels and glutathione peroxidase activity in the pre-frontal cortex in mice exposed to SD. Besides, the increase in hippocampal AChE activity induced by SD was overturned by SeBZF1. Lastly, cortical MAO-B activity was reestablished by SeBZF1 in mice that underwent SD. Based on the main findings of this study, it can be inferred that the compound SeBZF1 reverses the neurobehavioral alterations induced by sleep deprivation in male Swiss mice.

Abstract
BACKGROUND Evidence links major depressive disorder (MDD) with aging, but it's unclear if MDD accelerates aging and what factors mediate this transition. METHODS Two-sample Mendelian randomization (MR) analyses were applied to estimate the causal association between MDD and frailty index (FI), telomere length (TL), and appendicular lean mass (ALM) from available genome-wide association studies in populations of European ancestry. Furthermore, we conducted mediation MR analyses to assess the mediating effects of 31 lifestyle factors or diseases on the causal relationship between MDD and aging. RESULTS MDD was significantly causally associated with increased FI (βIVW = 0.23, 95 % CI = 0.18 to 0.28, p = 1.20 × 10-17), shorter TL (βIVW = -0.04, 95 % CI = -0.07 to -0.01, p = 0.01), and decreased ALM (βIVW = -0.07, 95 % CI = -0.11 to -0.03, p = 3.54 × 10-4). The mediation analysis through two-step MR revealed smoking initiation (9.09 %), hypertension (6.67 %) and heart failure (5.36 %) mediated the causal effect of MDD on FI. Additionally, alcohol use disorders and alcohol dependence on the causal relationship between MDD and TL were found to be 17.52 % and 17.13 % respectively. LIMITATIONS Confounding, statistical power, and Euro-centric focus limit generalization. CONCLUSION Overall, individuals with MDD may be at a higher risk of experiencing premature aging, and this risk is partially influenced by the pathways involving smoking, alcohol use, and cardiovascular health. It underscores the importance of early intervention and comprehensive health management in individuals with MDD to promote healthy aging and overall well-being.

Abstract
BACKGROUND Recent research has found a non-linear relationship between sleep duration and mental health/quality of life. However, it is currently unclear which age group is more affected by sleep duration and what the optimal sleep duration is for each age group. Additionally, the underlying mechanisms of the non-linear relationship between sleep duration and quality of life are not well understood. METHODS Therefore, this study utilized questionnaire data from 20,962 participants in the Chinese PBICR-2022 database(Cross-sectional study) and constructed models to analyze the relationship between sleep duration and quality of life. RESULTS The results showed that the optimal sleep duration varied among different age groups: approximately 9 h for adolescents, 8 h for early/middle-aged adults, and 7 h for older adults. Compared to other age groups, less or more sleep duration had a greater impact on mental health and quality of life in adolescents. Furthermore, mental health was found to play a significant mediating role between sleep duration and quality of life in both the less sleep group and the more sleep group, but this mediating effect was not significant among the older adults in the less sleep group, whereas the mediating effect of mental health was not significant among the middle-aged adults in the more sleep group. CONCLUSIONS This suggests that different age groups have different optimal sleep duration, and age may be an important factor influencing the relationship between sleep duration and mental health/quality of life, with mental health playing a mediating role in the relationship between sleep duration and quality of life.

Abstract
OBJECTIVE This study aimed to examine the association between past/current sleep duration and macro-/micro-structural brain outcomes and explore whether hypertension or social activity plays a role in such association. METHODS Within the UK Biobank, 40 436 dementia-free participants (age 40-70 years) underwent a baseline assessment followed by a brain magnetic resonance imaging (MRI) scan 9 years later. Past (baseline) and current (MRI scans) sleep duration (hours/day) were recorded and classified as short (≤5), intermediate (6-8), and long (≥9). Brain structural volumes and diffusion markers were assessed by MRI scans. RESULTS Compared with past intermediate sleep, past short sleep was related to smaller cortex volumes (standardized β [95 % CI]: -0.04 [-0.07, -0.02]) and lower regional fractional anisotropy (FA) (-0.08 [-0.13, -0.03]), while past long sleep was related to smaller regional subcortical volumes (standardized β: -0.04 to -0.07 for thalamus, accumbens, and hippocampus). Compared to current intermediate sleep, current short sleep was associated with smaller cortex volumes (-0.03 [-0.05, -0.01]), greater white matter hyperintensities (WMH) volumes (0.04 [0.01, 0.08]), and lower regional FA (-0.07 [-0.11, -0.02]). However, current long sleep was related to smaller total brain (-0.03 [-0.05, -0.02]), grey matter (-0.05 [-0.07, -0.03]), cortex (-0.05 [-0.07, -0.03]), regional subcortical volumes [standardized β: -0.05 to -0.09 for putamen, thalamus, hippocampus, and accumbens]), greater WMH volumes (0.06 [0.03, 0.09]), as well as lower regional FA (-0.05 [-0.09, -0.02]). The association between current long sleep duration and poor brain health was stronger among people with hypertension or low frequency of social activity (all Pinteraction <0.05). CONCLUSIONS Both past and current short/long sleep are associated with smaller brain volume and poorer white matter health in the brain, especially in individuals with hypertension and low frequency of social activity. Our findings highlight the need to maintain 6-8 h' sleep duration for healthy brain aging.

Abstract
Sleep deprivation (SD) has emerged as a critical concern impacting human health, leading to significant damage to the cardiovascular system. However, the underlying mechanisms are still unclear, and the development of targeted drugs is lagging. Here, we used mice to explore the effects of prolonged SD on cardiac structure and function. Echocardiography analysis revealed that cardiac function was significantly decreased in mice after five weeks of SD. Real-time quantitative PCR (RT-q-PCR) and Masson staining analysis showed that cardiac remodeling marker gene Anp (atrial natriuretic peptide) and fibrosis were increased, Elisa assay of serum showed that the levels of creatine kinase (CK), creatine kinase-MB (CK-MB), ANP, brain natriuretic peptide (BNP) and cardiac troponin T (cTn-T) were increased after SD, suggesting that cardiac remodeling and injury occurred. Transcript sequencing analysis indicated that genes involved in the regulation of calcium signaling pathway, dilated cardiomyopathy, and cardiac muscle contraction were changed after SD. Accordingly, Western blotting analysis demonstrated that the cardiac-contraction associated CaMKK2/AMPK/cTNI pathway was inhibited. Since our preliminary research has confirmed the vital role of Casein Kinase-2 -Interacting Protein-1 (CKIP-1, also known as PLEKHO1) in cardiac remodeling regulation. Here, we found the levels of the 3' untranslated region of Ckip-1 (Ckip-1 3'UTR) decreased, while the coding sequence of Ckip-1 (Ckip-1 CDS) remained unchanged after SD. Significantly, adenovirus-mediated overexpression of Ckip-1 3'UTR alleviated SD-induced cardiac dysfunction and remodeling by activating CaMKK2/AMPK/cTNI pathway, which proposed the therapeutic potential of Ckip-1 3'UTR in treating SD-induced heart disease.

Design setting participants This cross-sectional analysis used ordinary least squares regression within the UK Biobank database, assessing 26 820 participants aged 53-86 years, categorised into two cohorts: Cohort 1 (10 067 participants, 56% female; completed all four cognitive tests of Fluid Intelligence/reasoning, Pairs Matching, Reaction Time and Prospective Memory) and Cohort 2 (16 753 participants, 56% female; completed only two cognitive assessments of Pairs Matching and Reaction Time). Exposures Participant's self-reported sleep duration, chronotype and quality. Cognitive function was assessed through standardised computerised tests. The analysis was adjusted for demographic and comorbidity covariates. Main outcomes and measures Cognitive performance scores were evaluated against sleep parameters and health and lifestyle factors including sex, age, vascular and cardiac conditions, diabetes, alcohol intake, smoking habits and body mass index. Results The regression highlighted a positive association between normal sleep duration (7-9 hours) and cognitive scores in Cohort 1 (β=0.0567, 95% CI 0.0284 to 0.0851), while extended sleep duration negatively impacted scores across both cohorts (Cohort 1: β=-0.188, 95% CI -0.2938 to -0.0822; Cohort 2: β=-0.2619, 95% CI -0.3755 to -0.1482). Chronotype distinctions, particularly intermediate and evening types, were linked to superior cognitive function. Gender, age, angina, high blood pressure, diabetes, alcohol intake and smoking emerged as significant cognitive influencers. Conclusions and relevance The study delineates a multifaceted and nuanced relationship between sleep variables, health and lifestyle factors in determining cognitive outcomes. These findings highlight the vital role of sleep quality on cognitive health.

Abstract
Cross-sectional studies have demonstrated strong associations between physical frailty and depression. However, the evidence from prospective studies is limited. Here, we analyze data of 352,277 participants from UK Biobank with 12.25-year follow-up. Compared with non-frail individuals, pre-frail and frail individuals have increased risk for incident depression independent of many putative confounds. Altogether, pre-frail and frail individuals account for 20.58% and 13.16% of depression cases by population attributable fraction analyses. Higher risks are observed in males and individuals younger than 65 years than their counterparts. Mendelian randomization analyses support a potential causal effect of frailty on depression. Associations are also observed between inflammatory markers, brain volumes, and incident depression. Moreover, these regional brain volumes and three inflammatory markers-C-reactive protein, neutrophils, and leukocytes-significantly mediate associations between frailty and depression. Given the scarcity of curative treatment for depression and the high disease burden, identifying potential modifiable risk factors of depression, such as frailty, is needed.

Abstract
Background Negative emotions in college students are a significant factor affecting mental health, with suicide behaviors caused by negative emotions showing an annual increasing trend. Existing studies suggest that physical exercise is essential to alleviate negative feelings, yet the intrinsic mechanisms by which it affects negative emotions have not been fully revealed. Objective Negative emotions in college students represent a significant issue affecting mental health. This study investigates the relationship between physical exercise and negative emotions among college students, incorporating sleep quality and self-rated health (SRH) as mediators to analyze the pathway mechanism of how physical exercise affects students' negative emotions. Methods A cross-sectional study design was utilized, employing online questionnaires for investigation. The scales included the Physical Activity Rating Scale-3 (PARS-3), the Depression Anxiety Stress Scales-21 (DASS-21), the Pittsburgh Sleep Quality Index (PSQI), and the 12-Item Short Form Health Survey (SF-12), resulting in the collection of 30,475 valid questionnaires, with a validity rate of 91%. Chain mediation tests and Bootstrap methods were applied for effect analysis. Results The proportions of university students engaged in low, medium, and high levels of physical exercise were 77.6, 13.1, and 9.3%, respectively. The proportions of students experiencing "very severe" levels of stress, anxiety, and depression were 4.5, 10.9, and 3.6%, respectively. Physical exercise was significantly positively correlated with self-rated health (r = 0.194, p < 0.01), significantly negatively correlated with sleep quality (r = -0.035, p < 0.01), and significantly negatively correlated with stress, anxiety, and depression (r = -0.03, p < 0.01; r = -0.058, p < 0.01; r = -0.055, p < 0.01). Sleep quality was significantly negatively correlated with self-rated health (r = -0.242, p < 0.01). Mediation effect testing indicated that sleep quality and self-rated health partially mediated the relationship between physical exercise and negative emotions, with total effect, total direct effect, and total indirect effect values of -1.702, -0.426, and - 1.277, respectively. Conclusion College students primarily engage in low-intensity physical activity. Sleep quality and self-rated health mediate the impact of physical exercise on students' negative emotions. A certain level of physical activity can directly affect students' emotional states and indirectly influence their negative emotions via sleep and self-rated health. Regular engagement in physical activities primarily positively impacts emotional states by enhancing mood stability and overall emotional resilience.
