The experiments were categorized into the following groups: Transfection of vehicle in normoxia, transfection of vehicle in hypoxia, hypoxia-inducible factor-1α (HIF-1α) overexpression, HIF-1α knockdown, and HIF-1α overexpression plus HIF-1α knockdown in hypoxia. A: Cell proliferative capacity of peripheral blood mesenchymal stem cells was assessed by cell colony formation assays and crystal violet staining; B: Quantitative analysis of the number of cell clones in each group; C: Β-galactosidase staining was employed to examine cellular senescence of peripheral blood mesenchymal stem cells; D: Quantitative analysis of percentage of β-galactosidase positive cells (scale bars = 50 μm).
