Abstract
AIMS To investigate the therapeutic effect of drugs onchronic hepatitis B
virus infection.
METHODS Effects of rhIFN-αb and extracts of phyllanthus urinaria L. on
HBsAg production in PLC/PRF/5 human hepatoma cell line were in vestigated in
vitro.
RESULTS Decreased HBsAg production was regularly demonstrated 48h
after the treatment of cells with 50-250IU/mL of
rhIFN-αb and 2-4mg/mL phyllanthus urinaria L,
especially combining IFN with 10-5mol/L
Ara-A. Treatment of PLC/PRF/5 cells with hydrocortisone acetate (10-3mol/L)
and Ara-A(10-5mol/L) greatly stimulated HBsAg production. The
decrease of extracellular HBsAg excretion seemed to be due to inhibition of intracellular HBsAg formation.
CONCLUSIONS Interferon alfa, in combination with DNA synthesis inhibitors
may decrease the HBsAg formation in PLC/PRF/5 cells and a crudeextracts from
phyllanthus urinaria L. can also inhibit the HBsAg formation. This cell line is a convenient model for deeper studies of
chemotherapy of chronically HBV carriers.
